One of the best ways to speed your machine up to to uninstall all the software you don't use. If you have many icons in the system tray by the clock, then your PC is loading a lot of probably unneccesary software at boot time.
Other things like Symantec Antivirus can cause the machine to run slowly.
If you are using Vista, maybe consider using 'Sleep' rather than 'Shutdown'?
