Not really. You can block them from following you, but you can't prevent them from going to your twitter page and seeing it. As long as your account is public, anybody (whether they are logged in as twitter users or not) can see all of your tweets.
The best way around this, other than making your account private, is to use a twitter username that doesn't give your real name, and then don't let this one person find out about it.
Admittedly you would need your friends' help to keep the secret; and this still won't work if the person knows who your friends are (by looking at the list of people you follow now) - s/he can just look at their follows and figure it out.
