Twilio can currently connect with any POTs (plain old telephone) line in the US like your home/office phone or mobile phone.
Is there a specific 2.5G concern you had?

Twilio runs as service inside the network so your cell phone does not need Internet capabilities to connect to with Twilio. You can call Twilio or Twilio can call you using the good old telephone network.
While someone could use the Twilio REST API to initiate a call using a mobile phone, that call would originate from Twilio to another phone. i.e., you could have Twilio call your mobile phone but that call would route over the telephone network not the data network to ring your mobile.
