We are using VPN-Cubed vpcPLUS Free Edition manager on Amazon VPC for testing environment. Should I terminate and recreate a instance, routing every time?

The answer to your question depends on what you are trying to accomplish.
VPN-Cubed manager instances are meant to be up and running all the time, just like your regular router in your office or datacenter.
If for some reason your use case requires you to start and stop it often, you can either do it manually or automate this procedure. Using EC2 and VPC APIs you can automate starting a new instance and setting up routing, and using VPN-Cubed API you can automate loading your snapshot so that you get the same keyset and IPsec setup every time.
To stop the VPN-Cubed instance, you terminate the instance using EC2 API or EC2 tools.
Thanks for answer, but you forgot in your explanation deleting old route and creating new one in VPC.
Primary objective for me to use VPN-Cubed FREE Manager is VPC infrastructure testing, testing and again testing. I have to prepare infrastructure for QA team tests. They may start next week, may be during my holidays. In this case I need the instances to be ready to work only during the tests. I would request a stop instance future for testing environment. It ups to you.
I agree with you - VPN-Cubed manager has to be up non stop in production environment. In this case reasonable go to LITE and upper.
All operations in VPC and EC2, including adding and deleting of routes, can be performed with Amazon APIs.
A VPN-Cubed manager instance can't be stopped, because non-EBS-backed images in EC2 can't be stopped. We provide a snapshot capability which facilitates re-instantiating a new VPN-Cubed manager from an existing known state. These are VPN-Cubed snapshots, not S3 or EBS snapshots. You can read more about our snapshots in our documentation.
Alternatively, you could run VPN-Cubed manager all the time and start instances for your QA team on demand.
