Overall, a pretty comprehensive list.

FYC, but a couple seem to be pushing the boundaries a bit. There are plenty of high profile unlisted official options (in the employ of an espionage agency) like Dwayne Johnson as Bob Stone in Central Intelligence (2016), Charlize Theron as Lorraine Broughton in Atomic Blonde (2017) or Atomic Blonde 2, Steve Carell  as Maxwell Smart in Get Smart (2008) or real-life spy Ben Affleck as Tony Mendez in Argo (2012) that come to mind first. I would list those over these options that are more unofficial "spies" or contacts, unwitting agents, assets or useful idiots, such as Roger Thornhill, Severus Snape, Holly Martins and Nicholas Brody.

PS I wouldn't mind seeing these two on the list either

Harrison Ford
in Clear and Present Danger (1994) or

Ulrich Mühe
in Das Leben der Anderen (2006)

PSS I love Three Days of the Condor and it might be my favorite Redford spy. Redford has been a spy on more than a few occasions, but Robert Redford as Nathan Muir in Spy Game (2001) could be arguably his best character at spycraft.

Which spies would need spoilers? If Kevin Costner as Tom Farrell in No Way Out (1987). I would agree it would need a spoiler. But, if someone hasn't seen it after thirty years, I would think at some point you are relieved of the responsibility.

I have never seen One Flew Over The Cuckoo's Nest. I would not want some poll to have a spoiler in it.

Perhaps there are several kinds of "spy", but what stands out are the two categorizations "HUMINT" and "SIGINT", with the related condition of being either covert or overt, as well as the question of privilege level. For all cases, it is imperative that target of spying not be aware that he/she is being targeted, let alone how or by whom.  So, on one hand, the intelligence apparatus (or otherwise merely the spy master) could send somebody to develop a relationship with the target under false pretenses, and other hand, follow much more of a ghost approach that may involve blending in with a crowd, blending in with an inanimate environment, planting sensors, tampering with the environment to accomplish those things, observation via acoustic magnification, observation via telescopic means, or interception of telecommunications.
